10x Genomics is a single-cell library preparation system based on microfluidic technology, which can simultaneously obtain more than 1,000-80,000 single cells, prepare libraries compatible with Illumina platform, classify cell populations and analyze differences in gene expression between cell populations through sequencing of high-throughput single cell data. 10x Genomics single-cell expression analysis is an excellent method for studying tumor heterogeneity, immune cell population, and embryonic development with a wide range of cell types including tumor cells, immune cells, stem cells, nerve cells, germ cells, and embryonic cells.

Requirements

Sampling requirements
Cell isolationThe preparation of single cell suspensions is completed by any partner or Annoroad.
Sample typeCell suspension of fresh intact cells in vivo; frozen cell suspension; samples not suitable for fixation, staining, etc.
Total amount of sampleTotal amount >6*105 PCS; 2 copies of the same cell are prepared in parallel (one for inspection and one for experiment).
Sample concentration700-1200 pcs/μl.
Sample qualityCells should be free of clumps, with less fragments, survival rate >90%, cell diameter< 30 μm, and free of reverse transcription inhibitors and non-cellular nucleic acid molecules.
Cell cryopreservationThey should be stored in aliquots in the cell cryoprotectant: DMEM+20%FBS+10%DMSO.
Cell concentration of cryoprotectant1 million cells /ml; each 1 ml is dispensed into one cryovial.
